|a Sažetak: Stvaranje spore u stanici bakterije način je njezina preživlajvanja u nepovoljnim životnim uvjetima. Cijeli niz promjena do kojih dolazi pri ovoj pretvorbi nije još posve objašnjen, no poznato je da se peptidoglikan vegetativne bakterije, karakteristični gradivni element stanične stijenke, razlikuje od peptidoglikena spore. Ciklizacija muraminske kiseline, jednog od glavnih konstituenata peptidoglikana, u muramil-[delta-laktam nije nikada opažena kod vegetativnih bakterija, ali je karakteristična za peptidoglikan spora. Predmet istraživanja u ovoj radnji su spojevi iz reda muramil-[delta]-laktama peptidoglikana spora o kojima se, za razliku od spojeva karakterističnih za peptidoglika vegetativnih bakterija, nedovoljno zna.

|a Utjecaj zatvaranja 1,6-anhidro- [delta]-laktamskog prstena na konformaciju glukopiranoze muraminskog dijela peptidoglikana proučavan je na derivatima murmanske kiseline, 1,6-anhidromuraminske kiseline, muramil-[delta]-laktama i 1,6-anhidromuramil-[delta]-laktama metodama rendgenske strukturne analize, molekularne mehanike i dinamike. U disaharidnim derivatima pažnja je posvećena konformaciji oko glikozidne veze. Rezultati dobiveni rendgenskom strukturnom analizom i emprirjskim računalnim metodama uspoređeni su s rezultatima dobivenim pretraživanjem kristalografske baze podataka Cambridge structural Database i konformacijama istraživanih derivata prisutnim u otopini (mjerenje nuklearne magnetske rezonancije).

|a Abstract: In the case of invironmental conditions that are not suitable for life processes spore formation in bacteria serves as a startegy for survival. The multistage transformation process from bacterial vegetative cell into metabolically inactive spore has been subject of intense research activity in molecular biology but information about the changes by which the bacteria attains resistant state is still very scarce. However, it is known that the peptidoglycan of the spore, one of the constituents that build bacterial cell wall, has a composition different from that of the vegetative bacteria. Muramic acid closure in [delta]-lactam has never been observed in the peptidoglycans of the cell walls of vegetative bacteria, but characterizes the spore cell walls.

|a The aim of this work is to study the chemistry of muramic acid [delta]- lactam compounds to which no attention has been paid in contrast to the peptidoglycan type compounds that appear in vegetative bacteria which have been studied intensively. The influence of the closure of 1.6-anhydro-and-[delta]-lactam rings on the conformation of the muramic glucopyranose moiety was studied on the derivatives of muramic acid, 1,6-anhydromuramic acid, muramic acid [delta]-lactam and 1,6-anhydromuramic acid [delta]-lactam by X-ray structure analysis, molecular mechanics and dynamics. In the disaccharide derivatives possible conformations around glycosidic bond were investigated. The results obtained the discussed in comparison with the structural data extraced from the Camridge Structural Database and the data for the solution (nuclear magnetic resonance).
